The amount of dye reduced is proportional to the Vitamin C present. ๐Ÿงช Reagents: โ–ช DCPIP Solution โ–ช Oxalic Acid (as diluent) โ–ช Standard Ascorbic Acid Solution โš—๏ธ Procedure: โ— Take sample in conical flask. โ— Add 5 ml oxalic acid. โ— Titrate against DCPIP until light โ— pink color persists (30 seconds). โ— Repeat with standard. ๐Ÿงฎ Calculation: Vit C (mg/100ml) = (Sample titre / Standard titre) ร— Standard concentration โœ… Vitamin A (Retinol) Estimation ๐Ÿ”ฌ Principle: Vitamin A reacts with Trichloroacetic acid (TCA) or Antimony trichloride forming a blue-colored complex, measured colorimetrically. ๐Ÿงช Reagents: โ–ช TCA in chloroform โ–ช Vitamin A standard โ–ช Chloroform extract of sample โš—๏ธ Procedure: โ— Mix sample with TCA reagent. โ— Allow blue color to develop. โ— Measure absorbance at 620 nm. ๐Ÿงฎ Calculation: Compare absorbance with standard curve. โœ… Vitamin D (Calciferol) Estimation ๐Ÿ”ฌ Principle: Vitamin D absorbs UV light at 265 nm. Estimation done via spectrophotometer or HPLC. ๐Ÿงช Reagents: โ–ช Chloroform โ–ช Standard Vitamin D โ–ช UV Spectrophotometer โš—๏ธ Procedure: โ— Extract sample using chloroform. โ— Measure absorbance at 265 nm. โ— Compare with standard curve. ๐Ÿงฎ Note: HPLC provides better accuracy in clinical labs. โœ… Vitamin B1 (Thiamine) Estimation ๐Ÿ”ฌ Principle: Thiamine is oxidized and produces a fluorescent compound measured fluorometrically. ๐Ÿงช Reagents: โ–ช Potassium ferricyanide โ–ช Alkaline solution โ–ช Thiamine standard โš—๏ธ Procedure: โ— Oxidize sample with ferricyanide. โ— Measure fluorescence intensity. โ— Compare with standard. โœ… Vitamin B2 (Riboflavin) Estimation ๐Ÿ”ฌ Principle: Riboflavin is naturally fluorescent and detected under UV after acid hydrolysis. ๐Ÿงช Reagents: โ–ช๏ธ HCl (for hydrolysis) โ–ช๏ธ Riboflavin standard โš—๏ธ Procedure: ๐Ÿ”ธ Hydrolyze sample using acid. ๐Ÿ”ธ Neutralize & extract riboflavin. ๐Ÿ”ธ Measure fluorescence at 450 nm.
